DESCRIPTION OF VIRUSES

Subviral Agents: Viroids

Genus Pelamoviroid

Genus Pelamoviroid

Type Species

Peach latent mosaic viroid

(PLMVd)

Distinguishing Features

A circular ssRNA between 335 and 338 bases depending on isolates and sequence variants. The most stable secondary structure is a branched conformation in which neither five domains nor a central conserved region (CCR) can be distinguished (Fig. 6). It is insoluble in 2  M LiCl as is Chrysanthemum chlorotic mottle viroid (CChMVd) which also has a branched most stable secondary structure. Peach latent mosaic viroid (PLMVd) can form hammerhead structures in both polarity strands, and plus and minus monomeric PLMVd RNAs self-cleave in vitro as predicted by these structures, which are thermodynamically stable and most probably act as single hammerhead structures as in CChMVd but not in ASBVd (Fig. 3). Replication most probably occurs through a symmetric rolling-circle model by analogy with ASBVd (Fig. 3). Recorded in many peach-growing areas particularly in those raising American or Japanese cultivars: USA, Japan, China, and the Mediterranean basin. The only known natural host is peach but it can also be experimentally transmitted to peach hybrids. There are conflicting reports on whether or not PLMVd infects other Prunus species.

List of Species Demarcation Criteria in the Genus

An arbitrary level of less than 90% sequence similarity and distinct biological properties, particularly host range, currently separates species within genera.
